How do I read the tolerance test?

Try the peg in each hole. The smallest clearance where it slides in by hand is your sliding fit; the one before it is a press fit.

Should I re-run the test for each filament?

Yes — different materials and print temperatures shrink differently, so test each filament you use for fitted parts.
